Pediatric prosthetic users present unique challenges for device design and patient clinical care. Most children who use a prosthesis are born without a hand which is unlike most adult users who most typically lose a limb later in life. These children’s affected forearm muscles will have never actuated an intact hand, and this lifelong absence shapes muscle control in ways fundamentally different from adult amputees. Many children adapt early in life by developing compensatory strategies during activities of daily living and often feel no sense of loss but may feel different from their peers. My laboratory aims to understand how cells transduce mechanical signals and to use these insights to develop new therapies for disease and regeneration. In this talk, I will share our recent findings on the roles of mechanical forces in skeletal development and regeneration, our deep learning-enabled engineering of new optogenetic tools for the study of mechanobiology, and our discoveries of mechanobiology-inspired interventions for developmental disorders of the skeleton and of new cellular targets for promoting tissue regeneration. Lesions of the cortico-spinal tract (CST) and cortico-bulbar tract (CBT), resulting from stroke, traumatic brain injury (TBI), brain tumors, or neurodegenerative disorders, disrupt communication between the cortex and lower motor centers, leading to significant functional deficits in face, upper, or lower limb muscles. However, in most cases, the damage to the descending axons is incomplete offering a potential target for new therapies aimed at improving volitional muscle activation. The Cardiac Systems Simulation (CardSS) Lab in the Department of Bioengineering at the University of Washington in Seattle develops and applies new computational frameworks that help decipher mechanisms of heart disease and explore new frontiers of clinical treatment. In this lecture, CardSS Lab PI Dr. Pat Boyle will present a series of case studies on recent applications of this work.
