High-fidelity DNA sequencing of the single-strand origins of mutations Gilad D. Evrony, PhD Assistant Professor, Center for Human Genetics & Genomics NYU Grossman School of Medicine Why Attend this Seminar? Most mutations begin as a change to one of the two strands of the DNA double helix. However, these single-strand DNA changes have not been feasible to profile at single-molecule resolution. We have developed a DNA sequencing technology capable of profiling both single-strand and double-strand DNA changes with single-molecule fidelity. This includes both single-strand mismatches of canonical nucleotides and single-strand cytosine deamination damage. The revealed patterns of single-strand changes provide insight into the origins of mutations in several cancer predisposition syndromes. This work provides an initial framework for creating a catalogue of single-strand DNA damage and repair signatures that explain the known double-strand DNA mutation signatures.
